We present the first genetic maps of the sea cucumber ( Apostichopus japonicus ), constructed with an F1 pseudo-testcross strategy. The 37 amplified fragment length polymorphism (AFLP) primer combinations chosen identified 484 polymorphic markers. Of the 21 microsatellite primer pairs tested, 16 identified heterozygous loci in one or other parent, and six were fully informative, as they segregated in both parents. The female map comprised 163 loci, spread over 20 linkage groups (which equals the haploid chromosome number), and spanned 1522.0 cM, with a mean marker density of 9.3 cM. The equivalent figures for the male map were 162 loci, 21 linkage groups, 1276.9 and 7.9 cM. About 2.5% of the AFLP markers displayed segregation distortion and were not used for map construction. The estimated coverage of the genome was 84.8% for the female map and 83.4% for the male map. The maps generated will serve as a basis for the construction of a high-resolution genetic map and mapping of the functional genes and quantitative trait loci, which will then open the way for the application of a marker-assisted selection breeding strategy in this species.  相似文献

The newly introduced mosquito Aedes japonicus has expanded from its original range in Northeastern Asia to 29 US states (including Hawaii) plus Canada and northern Europe. Our objectives were to test an earlier hypothesis of multiple introductions of this species to the Northeastern US and evaluate putative temporal changes in genetic makeup. Using a panel of seven microsatellite loci, we confirmed the existence of two abundant genetic forms in specimens originally collected in 1999–2000 (FST value based on microsatellite data = 0.26) that matches the disjunctive distribution of mitochondrial haplotypes. To examine the distribution of the two genetic ‘types’ across Pennsylvania we created a fine‐scale genetic map of Ae. japonicus using 439 specimens collected from 54 Pennsylvania counties in 2002–2003. We also made direct comparisons between collections in 1999–2000 and new collections made in 2004–2005 obtained from the same areas in the northeastern US. We observed that the strong association between mtDNA haplotype and microsatellite signature seen in 1999–2000 had weakened significantly by 2002 across Pennsylvania, a trend continued to some extent in 2004–2005 in PA, NJ, and NY, indicating that once easily distinguishable separate introductions are merging. The two expanding genetic forms create a complex correlation between spatial and genetic distances. The existence of multiple introductions would be obscured without sampling early and across time with highly polymorphic molecular markers. Our results provide a high‐resolution analysis of the spatial and temporal dynamics of a newly introduced disease vector and argue that successive introductions may be a common pattern for invasive mosquitoes.  相似文献

The soft tick Argas japonicus mainly infests birds and can cause human dermatitis; however, no pathogen has been identified from this tick species in China. In the present study, the microbiota in A. japonicus collected from an epidemic community was explored, and some putative Rickettsia pathogens were further characterized. The results obtained indicated that bacteria in A. japonicus were mainly ascribed to the phyla Proteobacteria, Firmicutes and Actinobacteria. At the genus level, the male A. japonicus harboured more diverse bacteria than the females and nymphs. The bacteria Alcaligenes, Pseudomonas, Rickettsia and Staphylococcus were common in nymphs and adults. The abundance of bacteria belonging to the Rickettsia genus in females and males was 7.27% and 10.42%, respectively. Furthermore, the 16S rRNA gene of Rickettsia was amplified and sequenced, and phylogenetic analysis revealed that 13 sequences were clustered with the spotted fever group rickettsiae (Rickettsia heilongjiangensis and Rickettsia japonica) and three were clustered with Rickettsia limoniae, which suggested that the characterized Rickettsia in A. japonicus were novel putative pathogens and also that the residents were at considerable risk for infection by tick‐borne pathogens.  相似文献

目的:采用代谢组学方法研究麦冬对2型糖尿病(T2DM)小鼠内源性代谢物的影响。方法: 腹腔注射链脲佐菌素建立T2DM小鼠模型。连续4周灌胃给予麦冬水提物后,检测血清中糖尿病相关指标,且以超高效液相色谱-飞行时间质谱联用(UPLC-Q/TOF-MS)技术,研究麦冬对T2DM小鼠血清中代谢物质的影响,分析相关代谢通路。结果: 与正常对照组相比,模型组血清的空腹葡萄糖、总胆固醇、甘油三酯、低密度脂蛋白胆固醇含量显著升高,高密度脂蛋白胆固醇含量显著降低;灌胃给予麦冬水提物后,T2DM小鼠血清中以上指标均明显改善。代谢组学结果显示,正常组与模型组共有43个差异代谢物,富集于18条通路;麦冬提取物明显降低T2DM小鼠的甘油酸、丙二酸半醛和4-羟基苯基丙酮酸含量,差异代谢物富集于泛醌和其他萜类醌生物合成代谢等7条通路。结论: 麦冬水提物具有降低T2DM小鼠血糖和血脂的药效作用,且可能与泛醌和其他萜类醌生物合成等通路有关。  相似文献

Apostichopus japonicus is a kind of temperate sea cucumbers, known to aestivate when water temperature rises above 20°C to 24.5°C. In this study, we measured the effects of aestivation on the energy utilization (i.e., energy allocation in growth, feces discharge, respiration and excretion) of A. japonicus with two different body weights (134.0 g ± 3.5 g and 73.6 g ± 2.2 g) at water temperature from 10°C to 30°C with an interval of 5°C. Noticeable variation in the energy utilization of sea cucumbers was observed in this study. During the non-aestivation period, energy deposit in growth was lower and the energy loss in feces accounted for the majority of the feeding energy. Under the threshold temperature, the feeding energy reduced and the proportion of energy deposit in growth became negative. During aestivation, sea cucumbers discontinued feeding energy, resulting in weight loss. Our study suggested that the ecological implication of aestivation in this species could lead to a model of energy saving during the long-term hot period.  相似文献

We established a gene tagging population of the model legume Lotus japonicus using an endogenous long terminal repeat (LTR) retrotransposon Lotus Retrotransposon 1 (LORE1). The population was composed of 2450 plant lines, from which a total of 4532 flanking sequence tags of LORE1 were recovered by pyrosequencing. The two-dimensional arrangement of the plant population, together with the use of multiple identifier sequences in the primers used to amplify the flanking regions, made it possible to trace insertions back to the original plant lines. The large-scale detection of new LORE1 insertion sites revealed a preference for genic regions, especially in exons of protein-coding genes, which is an interesting feature to consider in the interaction between host genomes and chromoviruses, to which LORE1 belongs, a class of retrotransposon widely distributed among plants. Forward screening of the symbiotic mutants from the population succeeded to identify five symbiotic mutants of known genes. These data suggest that LORE1 is robust as a genetic tool.  相似文献

针对我国北方浅海筏式贝藻养殖系统自身污染输出导致的环境问题,以滤食性贝类生物沉积物和海藻粉的不同配比来模拟贝藻筏式养殖系统不同碳、氮负荷的颗粒自污染物,研究了刺参摄取这些颗粒物后的碳、氮收支,评估了其对碳和氮生源要素的生物清除潜力。结果表明,刺参对筏式贝藻养殖系统不同配比颗粒自污染物中的碳和氮具有较强的摄食能力,摄食率分别为35.77~181.18mgC·g-1·d-1和6.08~14.28mgN·g-1·d-1;颗粒自污染物中碳和氮的含量越高,其摄食碳、氮的能力越强。刺参摄取的碳以粪便碳形式排出居多(59.3%~97.1%),其次是呼吸消耗的碳(9.9%~37.3%),而用于生长的碳最少(-7.0%~6.1%);刺参摄取的氮主要用于排泄消耗为主(53.1%~63.1%),粪便氮次之(27.7%~39.2%),用于生长的氮最少(-2.3%~16.7%)。通过建立的碳、氮收支方程,估算出刺参对筏式贝藻养殖系统不同碳、氮负荷自污染物的生物清除效率(SE)分别为0.83~4.57mgC·g-1·d-1和0.28~0.76mgN·g-1·d-1;而且其清除效率随着颗粒自污染物中碳、氮含量的升高而增大,呈明显的正相关关系;清除效率和碳(C)、氮(N)含量之间回归关系可用SEC=0.7368+14.9488C和SEN=0.2281+0.2202N来描述。  相似文献

随着刺参养殖业的迅速发展,刺参池塘养殖暴露出诸多问题,如水体氨态氮含量高、病害严重、单位产量下降等.传统药物弊端较多,微生物方法成为降解水体中氨态氮含量和预防病害发生的新的有效途径.综合阐述了刺参池塘养殖过程中氨态氮对刺参的危害和微生物降解氨态氮的利弊,并介绍了水体中氨态氮高效降解菌株的筛选以及现有微生态制剂存在的问题,对微生物方法降解刺参养殖池塘氨态氮的前景进行了展望.  相似文献
